Comparative Studies on the Hibiscus syriacus and Its Allied Species Based on RAPD Analysis

Abstract

|
The phylogenetic relationships between Hibiscus syriacus (Malvaceae) and its allied species were investigated by RAPD (Randomly Amplified Polymorphic DNA) method. Twelve primers out of 100 primers (10-mer) screened gave rise to complete polymorphism (100%) in all of the tested, species, producing 233 randomly amplified DNA fragments. Twenty seven species were classified into 6 sectional groups by UPGMA clustering analysis. Section Bombycella including H. syriacus, H. paramutabilis, and H. sinosyriacus showed the most distinct separation from its allied sectional groups at the 0.68 of similarity index of RAPDs. This grouping was generally in accordance with Hochreutiner¢¥s (1900) sectional boundaries based on morphological studies. Furthermore, this study showed that RAPD technique could be used to solve problems in relation with the assignment of a species to a particular section. This study was comparatively discussed with previous other¢¥s sectional boundaries based on morphological characteristics.
